3D ORGAN IMAGING

Validation of nuclear medicine tracer

NUCLEAR TRACER VALIDATION IN GLIOBLASTOMA MODEL

Aim:  Study of the distribution of a radiolabeled tracer in a glioblastoma tumor. Current methods using autoradiography do not allow complete 3D exploration due to challenges in 2D serial section registration and the lack of anatomical information. This study presents a multimodal approach where tumor and vessel imaging using the Kratoscope can be registered to the autoradiography imaging in order to model the activity distribution as a function of time.

Approach: Animals were injected with the nuclear radionuclide and subsequently with a vascular contrast agent. Autofluorescence imaging was performed on a fresh frozen tumor. 2D sections were collected and autoradiography acquisitions and subsequent HES staining were performed.

Figure: A. Autofluorescence signal of the tumor was imaged using the Kratoscope. Vessels appear in dark as a result of the injected vascular contrast agent, and tumor vasculature can be segmented and 3D reconstructed (D). B. Autoradiography measurement (BeaQuant) reveal nuclear activity in the corresponding 2D section (B), and HES staining shows cellular organisation and composition of the tissue (C). Registration of these imaging modalities (E) reveal regions where high nuclear activity correlate with the presence of vessels (red circle, D), and will be further studied to model the activity distribution as a function of time (F).

Result: 3D reconstruction of the tumor and corresponding tumor vasculature. Co-registration with autoradiography and histology to assess activity distribution in the vicinity of the vessels.
